SAR1A is a key member of the small GTPase superfamily, playing a crucial role in the export of cargo from the Endoplasmic Reticulum (ER). This protein serves as a molecular switch, controlling interactions between proteins and lipids that determine the formation of COPII vesicle coats for ER cargo export. SAR1A interacts with a membrane-bound exchange factor called Sec12 when it is in its GDP-bound state, exchanging its GDP for GTP. Additionally, SAR1A is involved in transporting materials from the ER to the Golgi apparatus. It is necessary for maintaining the localization of SEC16A, a protein found on the ER membrane, by preventing its dissociation. The assembly of SEC16A on the ER membrane by SAR1A-GTP forms a structured scaffold that defines endoplasmic reticulum exit sites.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
