S100b is a member of the S100 protein family that primarily exists as dimers. These proteins are involved in various cellular processes and are found in the cytoplasm or nucleus of many different cell types. S100b specifically is expressed in cells of the central nervous system, glial cells, and certain peripheral cells. It is located on chromosome 21q22.3, while the other S100 genes are clustered on chromosome 1q21. Determining the levels of S100b in serum can help monitor brain injury and melanoma. S100b proteins play roles in neurite extension, melanoma cell proliferation, calcium flux stimulation, PKC-mediated phosphorylation inhibition, astrocytosis and axonal proliferation, and microtubule assembly inhibition. Altered expression of the S100b gene and chromosomal rearrangements are associated with several neurological, neoplastic, and other diseases like Alzheimer's disease, Down's syndrome, epilepsy, amyotrophic lateral sclerosis, melanoma, and type I diabetes.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
