S100A8 is a calcium-binding protein belonging to the S100 family of proteins, which contain 2 EF-hand calcium-binding motifs. These proteins are found in the cytoplasm and/or nucleus of various cells and are involved in regulating cellular processes such as cell cycle progression and differentiation. S100A8 has been shown to inhibit casein kinase and act as a cytokine. Its expression has been found to be altered in cystic fibrosis disease. Moreover, S100A8 exhibits antimicrobial activity against bacteria and fungi, making it essential for defense against pathogenic bacteria. It also up-regulates the transcription of genes controlled by NF-kappa-B and contributes to the development of endotoxic shock in response to bacterial lipopolysaccharide. Additionally, S100A8 promotes tubulin polymerization and facilitates migration of phagocytes and infiltration of granulocytes at wound sites. It acts as a pro-inflammatory mediator in both acute and chronic inflammation and up-regulates the release of IL8 and the cell-surface expression of ICAM1.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
