S100A8 is a member of the S100 protein family, which contains 2 EF-hand calcium-binding motifs. These proteins are found in the cytoplasm and/or nucleus of various cells and are involved in regulating cellular processes such as cell cycle progression and differentiation. S100A8 serves as an inhibitor of casein kinase and functions as a cytokine. Altered expression of S100A8 is associated with cystic fibrosis. It acts as a calcium-binding protein with antimicrobial activity against bacteria and fungi. S100A8 is critical for defending against invasion by pathogenic bacteria. It enhances the transcription of genes regulated by NF-kappa-B and contributes to the development of endotoxic shock caused by bacterial lipopolysaccharide. Additionally, S100A8 promotes tubulin polymerization and facilitates the migration and infiltration of immune cells at wound sites. It acts as a pro-inflammatory mediator in both acute and chronic inflammation, increasing the release of IL8 and the expression of ICAM1 on the cell surface.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
