Regulation of nuclear pre-mRNA domain-containing protein 1B (RPRD1B) interacts with phosphorylated C-terminal heptapeptide repeat domain (CTD) of POLR2A, participating in CTD dephosphorylation. RPRD1B enhances RNA polymerase II binding to the CCDN1 promoter and the termination region prior to the poly-A site, but decreases binding after the poly-A site. It inhibits RNA polymerase II from reading through the 3' end termination site, potentially leading to a chromatin loop formation and drawing it back to the promoter. RPRD1B also boosts transcription of various cell cycle-related genes (CDK2, CDK4, CDK6, cyclin-E), while not affecting CDKN1A, CDKN1B, or cyclin-A expression. Additionally, RPRD1B promotes cell proliferation.
