The ribosomal phosphoproteins (P proteins) are found in the large ribosomal subunit and are located in the cytoplasm. There are three types of P proteins: P0 (molecular weight 35 kDa), P1 (19 kDa), and P2 (17 kDa). RPLP0 is a ribosomal protein that belongs to the L10P family and is part of the 60S subunit. It is a neutral phosphoprotein that has a similar C-terminal end as the acidic ribosomal phosphoproteins P1 and P2. P0 interacts with P1 and P2 to form a pentameric complex. Multiple processed pseudogenes of RPLP0 can be found throughout the genome, which is common for genes encoding ribosomal proteins. In systemic lupus erythematosus (SLE) patients, 10% have autoantibodies against ribosomal P proteins. If these antibodies are present without other typical SLE-associated autoantibodies, they may explain some cases of ANA-negative lupus. It has been observed that SLE patients with anti-ribosomal P autoantibodies often have involvement of the central nervous system, suggesting that these antibodies could be used as markers for this condition.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
