The human gene located on chromosome 21, known as RCAN1, has the ability to interact with calcineurin A and hinder calcineurin-dependent signaling pathways. This interaction may have an impact on the development of the central nervous system. RCAN1 is found in the specific region of interest for the Down syndrome phenotype and is observed to be overexpressed in the brains of Down syndrome fetuses. Overexpression of this gene over a prolonged period may contribute to the formation of neurofibrillary tangles, which are also associated with Alzheimer's disease. Additionally, three different versions of the gene, referred to as transcript variants, have been discovered, each encoding a distinct isoform. Another gene called DSCR1 has a similar role in inhibiting calcineurin-dependent transcriptional responses by binding to the catalytic domain of calcineurin A. Like RCAN1, DSCR1's action may also affect the development of the central nervous system.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
