RAET1E is a member of the MHC class I family, which consists of MHC class I-related genes located on chromosome 6q24.2-q25.3. Unlike other RAET1 proteins, RAET1E and RAET1G proteins have type I membrane-spanning sequences at their C termini instead of glycosylphosphatidylinositol anchor sequences. RAET1E binds to the NKG2D receptor, which is expressed on various immune cells and is involved in both innate and adaptive immune responses. This interaction allows RAET1E to send signals to NK cells and promote tumor immune surveillance by stimulating the growth of anti-tumor cytotoxic lymphocytes.
