PTTG1, or Securin, plays a key role in cell division by regulating the separation of sister chromatids. It is involved in chromosome stability, the p53/TP53 pathway, and DNA repair. PTTG1 has two additional roles: facilitating the transport of separase to the nucleus and inhibiting its catalytic activity. It is ubiquitinated by the APC and degraded by the Proteasome, which releases separase. During mitosis, PTTG1 blocks Separase/ESPL1 function to prevent the cleavage of the cohesin complex and the subsequent segregation of chromosomes. However, PTTG1 is also required to activate ESPL1. At the beginning of anaphase, PTTG1 is ubiquitinated, leading to its destruction and the liberation of ESPL1. PTTG1 contains two PXXP motifs necessary for its transforming and tumorigenic activities, as well as its stimulation of basic fibroblast growth factor expression. It also contains a D box essential for its degradation by the APC. The acidic C-terminal region of Securin can serve as a transactivation domain. Although primarily cytosolic, PTTG1 is partially localized in the nucleus. It is highly expressed in various tumors and exhibits transforming and tumorigenic activities in vitro and in vivo. Additionally, PTTG1 negatively regulates the transcriptional and apoptotic activity of TP53.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
