PSMB10 is part of the proteasome B-type family (T1B family), specifically a 20S core beta subunit. The proteasome is a complex enzyme with a highly organized ring-shaped 20S core structure. This structure consists of four rings composed of 28 different subunits: two rings of seven alpha subunits and two rings of seven beta subunits. Proteasomes are abundant in eukaryotic cells and break down peptides in a process that requires ATP and ubiquitin, occurring outside of lysosomes. One important role of a modified proteasome called the immunoproteasome is to process class I MHC peptides. PSMB10 gene expression is stimulated by INFg and it replaces catalytic subunit 2 (proteasome beta 7 subunit) in the immunoproteasome.

Storage Instruction

Store at 4°C if entire vial will be used within 2-4 weeks. Store, frozen at -20°C for longer periods of time. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Avoid multiple freeze-thaw cycles.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
