The recombinant Protein A/G fusion protein is a combination of the IgG binding domains from Protein A and Protein G. It has four Fc binding domains from Protein A and two from Protein G, resulting in a total mass of 50.4 kDa. Protein A/G has a lower pH binding dependency compared to Protein A, but it possesses the combined properties of both Protein A and G. It is capable of binding to all subclasses of human IgG, making it useful for purifying polyclonal or monoclonal IgG antibodies when their subclasses are unknown. Additionally, Protein A/G binds to IgA, IgE, IgM, and IgD. When it comes to mouse IgG, Protein A/G can bind to all subclasses except for mouse IgA, IgM, and serum albumin. This makes it suitable for purifying and detecting mouse monoclonal IgG antibodies without interference from IgA, IgM, and serum albumin. Furthermore, chimeric Protein A/G exhibits a stronger affinity toward mouse monoclonal antibodies compared to Protein A or Protein G alone. It has also been utilized in the purification of macaque IgG.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
