SDF-1, also known as CXCL12, is a small cytokine belonging to the chemokine family. It is produced in two forms, SDF-1α/CXCL12a and SDF-1β/CXCL12b, through alternate splicing of the same gene. CXCL12 is a CXC chemokine with four conserved cysteines, two of which form disulfide bonds. It plays a key role in directing the migration of hematopoietic cells during embryonic development. Knock-out mice for the CXCL12 gene did not survive long after birth. CXCL12 is also involved in altering the electrophysiology of neurons. It is expressed in various tissues in mice, such as the brain, thymus, and spleen. The receptor for CXCL12 is CXCR4, previously known as fusin, although recent studies suggest CXCR7 may also bind to CXCL12. The gene for CXCL12 is located on human chromosome 10 and shows high sequence identity in both humans and mice.

The sequence of the first five N-terminal amino acids was determined and was found to be Lys-Pro-Val-Ser-Leu.

Storage Instruction

Lyophilized SDF-1a although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution CXCL12 should be stored at 4°C between 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

Sample Prep Instruction

It is recommended to reconstitute the lyophilized Stromal Cell-Derived Factor-1a in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
