S100b is a type of protein that belongs to the S100 family. This family consists of calcium-binding proteins called EF-hand proteins, which are usually found as dimers made up of 20 individual S100 monomers. S100B homodimers are present in cells of the central nervous system, glial cells, and certain peripheral cells like Schwann cells, melanocytes, adipocytes, and chondrocytes. These proteins are located either in the cytoplasm or the nucleus of various cells and are involved in regulating cellular processes such as cell cycle progression and differentiation. S100b is one of the 13 members of the S100 gene family, which are found together on chromosome 1q21. However, S100b itself is located at 21q22.3. Measurement of S100B levels in the blood is used to assess brain injury severity and malignant melanoma progression. S100b proteins have various functions, including promoting neurite extension, stimulating melanoma cell growth, modulating calcium flow, inhibiting a specific type of protein phosphorylation, promoting astrocyte and axonal growth, and disrupting microtubule assembly. Changes in the structure and expression of the S100b gene are associated with neurological disorders, cancerous conditions, and other diseases like Alzheimer's, Down's syndrome, epilepsy, amyotrophic lateral sclerosis, melanoma, and type I diabetes.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
