MMP-3, also known as Stromelysin-1 or Transin-1, is an enzyme that breaks down natural collagen at physiological pH and temperature. It is involved in degrading the intervertebral nucleus pulposus and annulus fibrosus of herniated lumbar intervertebral discs. MMP-3 targets various components of the extracellular matrix such as proteoglycan, laminin, fibronectin, gelatin, and collagen types III, IV, and IX. Additionally, MMP-3 can activate other matrix metalloproteinases like pro-MMP-9 and pro-MMP-8, and enhance the activity of plasmin-activated MMP-1. MMP-3 is initially secreted as an inactive form and becomes active through the action of various proteinases such as plasmin, trypsin, chymotrypsin, cathepsin G, or human neutrophil elastase. It has also been shown to activate the precursor of interleukin-1 beta.

The activity was measured by its ability to cleave the fluorogenic peptide substrate, Mca-RPKPVE-Nval-WRK(Dnp)-NH2. The specific activity is > 150 pmoles/min/µg.Recombinant Human MMP-3 protein pro form needs to be activated with Chymotrypsin.Activation Protocol:1. Dilute MMP3 to 20µg/ml in the Assay Buffer: 50mM Tris, 10mM CaCl2, 150mM NaCl, 0.05% (w/v) and Brij 35, pH 7.5.2. Activate MMP3 by adding Chymotrypsin(Sigma, Catalog#C­3142,1mg/ml stock in 1mM HCl) to a final concentration of 5ug/ml.3. Incubate at 37°C for 30 minutes.4. Stop activation with 2mM PMSF. Pre-warm the PMSF to 37°C prior to adding to sample.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
