MMP-3, also known as Stromelysin-1 or Transin-1, is an enzyme that hydrolyzes natural collagen at physiological pH and temperature. It is involved in breaking down the intervertebral nucleus pulposus and annulus fibrosus of Herniated Lumbar Intervertebral Disk. MMP-3 targets various components of the extracellular matrix, such as proteoglycan, laminin, fibronectin, gelatin, and collagen types III, IV, and IX. Additionally, MMP-3 is known to activate pro-MMP-9 and pro-MMP-8, and superactivate plasmin activated MMP-1. This enzyme is initially secreted as a latent proenzyme and can be activated by various proteinases such as plasmin, trypsin, chymotrypsin, cathepsin G, or human neutrophil elastase. MMP-3 has also been shown to activate the precursor of IL1-beta.

Recombinant Human Matrix Metalloproteinase-3 (MMP-3) is a member of the matrix metalloproteinase family, which plays a key role in the breakdown of extracellular matrix components. MMP-3 is involved in tissue remodeling and repair processes, as well as in various pathological conditions such as arthritis and cancer. With a molecular weight of approximately 57 kDa, MMP-3 is a zinc-dependent endopeptidase that cleaves a wide range of substrates, including collagen, proteoglycans, and cytokines. Its activity is tightly regulated to maintain tissue homeostasis and prevent excessive degradation of the extracellular matrix.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
