M2 autoantigen is an important marker for antimitochondrial autoantibodies (AMA) found in patients with primary biliary cirrhosis (PBC), an autoimmune liver disease that causes damage to bile ducts within the liver. The M2 antigen consists of at least three different proteins, similar to the E2 subunits of various mitochondrial dehydrogenase complexes. These complexes are responsible for the oxidative decarboxylation of alpha-keto-acid substrates and contain a prosthetic lipoamide group. They are located in the inner membrane of the mitochondria. The main reactivity of AMA positive PBC sera is against PDC-E2, but many patients also show reactivity against OGDC-E2 and/or BCOADC-E2. Some patients may only react with OGDC-E2 and/or BCOADC-E2 and not PDC-E2. These patients may be missed by assays that primarily use M2 antigen preparations derived from natural sources that mainly contain PDC-E2.

Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
