Heparanase is an enzyme that breaks down heparan sulfate side chains of heparan sulfate proteoglycans (HSPGs) in the extracellular matrix. This process is important for the degradation of the extracellular matrix, facilitating the movement of tumor cells and inflammatory leukocytes. As heparanase degrades heparan sulfate, it releases growth factors and cytokines that promote cell proliferation and chemotaxis. Heparanase is composed of two subunits, a 50 kDa subunit with the active site and an 8 kDa subunit. It is initially produced as a 65 kDa precursor and undergoes proteolytic processing to become active. Heparanase is highly expressed in myeloid leukocytes, platelets, and human placenta. In various types of primary tumors, human heparanase is upregulated and is associated with increased invasiveness, vascularity, and poor survival rate.

The specific activity of Heparanase Active Enzyme in-house standard is about 0.7 Units (1 unit = 1 μmole of reducing ends of heparan sulfate substrate formed per minute per mg Heparanase Active Enzyme at 37°C). The enzymatic activity of each Heparanase Active Enzyme batch is comparable to the standard as determined by activity assay in which immobilized heparan, released due to heparanase activity, is quantified colorimetrically. Recommended reaction buffer: 20 mM Citrate Phosphate buffer, pH 5.4; 50mM NaCl; 1mM CaCl2.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
