Glutathione peroxidase 1 (GPX1) is a crucial member of the glutathione peroxidase family, which consists of 8 identified glutathione peroxidases (Gpx1-8) in humans. This enzyme plays a key role in detoxifying hydrogen peroxide and is considered one of the most important antioxidant enzymes in humans. GPX1 is part of the enzymatic antioxidant defense system, protecting DNA, proteins, and lipids from oxidative damage by neutralizing hydrogen and lipid peroxides, which can potentially contribute to the development of prostate cancer. GPX1 is one of the few proteins in higher vertebrates that contains selenocysteine at its active site, coded by the TGA stop codon. Additionally, GPX1 features a polyalanine sequence polymorphism in the N-terminal region, with 3 alleles containing 5, 6, or 7 alanine (ALA) repeats in this sequence. The presence of 5 ALA repeats in the allele has been significantly associated with an increased risk of breast cancer.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
