Glycogen phosphorylase is an enzyme that breaks down glycogen into glucose subunits, leaving behind glucose-1-phosphate. For metabolism, glucose-1-phosphate must be converted to glucose-6-phosphate by phosphoglucomutase. Glycogen phosphorylase can only work on linear chains of glycogen and will stop when encountering a 1-6 branch. Debranching enzyme is needed to straighten out the chain, and alpha 1-6 glucosidase enzyme is required to break the remaining 1-6 residue to allow glycogen phosphorylase to continue. Phosphoprotein phosphatase (PP-1) inactivates glycogen phosphorylase in response to insulin stimulation to prevent glycogen breakdown. Glycogen phosphorylase BB (GPBB) is a sensitive marker for diagnosing acute myocardial infarction (AMI) within a few hours of chest pain onset. GPBB levels can also be elevated in patients with unstable angina or undergoing coronary artery bypass grafting surgery.

1. Thaw the vial on ice, spin briefly

Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
