Filamin is a large actin crosslinking protein that helps stabilize the 3D cortical actin network. It consists of an actin binding domain at the N-terminus, followed by a C-terminal rod domain consisting of repeat segments. The rod domain contains approximately 100 residues per repeat and adopts an immunoglobulin-like fold. This type of fold is found in many proteins and is responsible for protein-protein interactions. Mammalian filamin, also known as actin-binding protein (ABP), forms orthogonal actin filament networks in the cortical cytoplasm and anchors membrane proteins to the actin cytoskeleton. It interacts with multiple proteins, including transmembrane receptors, second messenger-associated proteins, protein kinases, phosphatases, and cytoskeletal proteins. These interactions depend on the repeat elements in the rod domain.
