The low affinity immunoglobulin gamma Fc region receptor III-A (FCGR3A) plays a role in removing antigen-antibody complexes from the circulation and carrying out other antibody-dependent immune responses. FCGR3A binds to the gamma subunit of Fc epsilon. The receptor is found on natural killer (NK) cells, where it is anchored to the cell membrane by a transmembrane peptide. FCGR3B, on the other hand, is found on polymorphonuclear neutrophils (PMN) and is anchored by a phosphatidylinositol (PI) linkage. FCGR3A is also found on macrophages, a subset of T-cells, immature thymocytes, and placental trophoblasts. It is involved in mediating antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis. Mutations in the FCGR3A gene are associated with an increased susceptibility to recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.

Recombinant Human CD16a, also known as FcγRIIIa, is a transmembrane glycoprotein belonging to the immunoglobulin superfamily. With a molecular weight of approximately 50-70 kDa, CD16a plays a significant role in mediating antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis. As a member of the Fcγ receptor family, CD16a binds to the Fc region of IgG antibodies, facilitating the activation of immune effector cells such as natural killer cells, macrophages, and neutrophils. This interaction triggers a cascade of signaling events, ultimately leading to the elimination of target cells.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
