Human Factor IX, also known as Christmas-Factor, is a glycoprotein synthesized in the liver. It belongs to the serine proteases system and is part of the S1 peptidase family. Lack of Factor-IX leads to Hemophilia-B or Christmas Disease. The N-terminus region of Factor-IX contains 12xGla residues that assist in the calcium-dependent binding of Factor-IX to the phospholipid surface. It can be activated by either factor XIa or the factor VIIa/tissue factor/phospholipid complex. This activation results in the formation of intermediate IXa, which is then converted to the fully active form IXab. Coagulation factor IX binds initially to exosites on the factor XIa heavy chain and then interacts at the active site, leading to bond cleavage. Interacting with the erythrocyte membrane, Factor-IX activation triggers intrinsic coagulation. Chaperones and lectins work together to ensure proper folding of Factor-IX and retention of mutant molecules. Human Factor IX, activated through either the Contact or Tissue Factor Pathway, activates Factor X to Xa.

Sample Prep Instruction

It is recommended to reconstitute the lyophilized 100U Factor-IX in sterile 100µl of 18MΩ-cm H2O, which can then be further diluted to other aqueous solutions.
