The antigen ESAT6, derived from the shot-term-culture filtrate (ST-CF) of Mycobacterium, is easily detectable in tuberculosis patients. A group of genes code for the secretory apparatus responsible for exporting ESAT-6 and related proteins, which are potent T-cell antigens. These genes are also recognized strongly by T cells, suggesting that ESAT-6 systems can be seen as immunogenicity islands. There is growing evidence that these genes are under selective pressure from the host's immune system. Most tuberculosis patients (over 90%) have detectable epitopes of this antigen in their serum. Efforts to create a vaccine using ESAT-6 have shown that the optimization of adjuvants, such as dioctadecylammonium bromide and monophosphoryllipide, is crucial. ESAT-6 is also promising as a diagnostic tool for distinguishing between mycobacterial infection and BCG vaccination. The main applications of ESAT-6 are in antibody production and the development of tuberculosis test systems. ESAT6 is secreted as a complex with cell filtrate protein 10 (CFP10) by replicating Mycobacterium tuberculosis complex (MTC).

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
