Mycobacterium antigen ESAT-6 is easily detectable in tuberculosis patients and has been isolated from low molecular weight fractions of the shot-term-culture filtrate (ST-CF). The export of ESAT-6 requires specific genes that code for proteins recognized by T cells. These genes are under selective pressure from the host's immune system. ESAT-6 contains epitopes detectable in the serum of over 90% of tuberculosis patients. When developing a vaccine based on ESAT-6, optimizing the adjuvant is crucial, particularly by using a combination of dioctadecylammonium bromide and monophosphoryllipide. ESAT-6 has shown promise as a diagnostic tool to differentiate between mycobacterial infection and BCG vaccination. The main applications of ESAT-6 are in antibody production and in developing test systems for tuberculosis diagnosis.

Recombinant Early Secretory Target Mycobacterium Tuberculosis (ESAT-6) is a protein primarily found in the pathogenic bacterium Mycobacterium tuberculosis. Belonging to the ESAT-6 family, this protein plays a significant role in the early stages of tuberculosis infection. With a molecular weight of approximately 6 kDa, ESAT-6 is involved in the modulation of host immune responses and the establishment of a successful infection. Through its secretion system, ESAT-6 aids in the evasion of the host immune system, allowing for the survival and proliferation of M. tuberculosis within the host.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
