HER-2/neu is a gene that codes for an orphan receptor tyrosine kinase known as erbB-2. This receptor is always active and forms dimers, and excessive expression can lead to cancer. A transcript of HER-2 called herstatin, which contains intron 8, has been identified. herstatin mRNA is found in normal fetal kidney and liver, but at lower levels compared to p185HER-2 mRNA in carcinoma cells with an amplified HER-2 gene. herstatin appears to inhibit p185HER-2 by disrupting dimers, reducing tyrosine phosphorylation, and inhibiting the growth of transformed cells that overexpress HER-2 independently of anchorage.
