ADAM10 is a member of the ADAM family of cell surface proteins. These proteins have a unique structure that includes both adhesive and protease domains. ADAM10 plays a role in cleaving various proteins, such as TNF-alpha and E-cadherin. For example, it cleaves the membrane-bound precursor of TNF-alpha to produce its mature soluble form by cleaving at position 76-ala-|-val-77. ADAM10 is also responsible for the proteolytic release of other cell-surface proteins, including heparin-binding epidermal growth-like factor and ephrin-a2. It is involved in the cleavage of amyloid precursor protein as both a constitutive and regulated alpha-secretase. ADAM10 is also involved in the normal cleavage of cellular prion protein, the adhesion molecule L1, and in the release of membrane vesicles, suggesting a vesicle-based protease activity. Additionally, ADAM10 controls the proteolytic processing of notch and mediates lateral inhibition during neurogenesis.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
