DKK1, a member of the dickkopf family, is a secreted protein that plays a role in embryonic development by inhibiting the WNT signaling pathway. Elevated levels of DKK1 in bone marrow plasma and peripheral blood are associated with osteolytic bone lesions in patients with multiple myeloma. DKK1 induction is also involved in the pathological cascade triggered by beta-amyloid and contributes to the development of tau phosphorylation. Additionally, DKK1 reduces the differentiation of osteoblastic precursor cells and is crucial in multiple myeloma bone disease. In contrast, DKK1 expression is reduced in human colon tumors, suggesting that it acts as a tumor suppressor gene in this neoplasia. The absence of DKK1 induction leads to downregulation of the Wnt/beta-catenin pathway, which is a mechanism missing in colon cancer.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
