DCTN2 is a component of dynactin, a macromolecular complex including 10-11 subunits of varying sizes (22-150 kDa). Dynactin binds to microtubules and cytoplasmic dynein and has various cellular functions such as transport from the ER to the Golgi apparatus, movement of lysosomes and endosomes, spindle formation, chromosome movement, nuclear positioning, and axonogenesis. DCTN2 is found in 4-5 copies per dynactin molecule and consists of 3 alpha-helical coiled-coil domains that facilitate its interaction with other dynactin subunits. DCTN2 directly interacts with the largest dynactin subunit (p150) and stabilizes its position. DCTN2 regulates the binding of cytoplasmic dynein to an organelle and contributes to chromosome alignment and spindle organization during mitosis. Additionally, DCTN2 plays a part in anchoring microtubules to centrosomes and is involved in synapse formation during brain development.
