Dystroglycan 1 (DAG1) is a component of the dystrophin-glycoprotein complex that connects the subsarcolemmal cytoskeleton to the extracellular matrix through its binding to laminin. The N-terminal domain of alpha-dystroglycan is found in the cerebrospinal fluid. The impact of DAG1 on the nervous system is not well understood. The dystroglycan complex is present in various tissues and plays a role in processes such as assembling laminin and basement membranes, maintaining sarcolemmal stability, promoting cell survival, facilitating peripheral nerve myelination, shaping nodal structure, promoting cell migration, and facilitating epithelial polarization. DAG1 is a possible candidate gene for mutations in autosomal recessive muscular dystrophies. In Duchenne muscular dystrophy, there is a significant reduction of DAG1 levels, which weakens the connection between the sarcolemma and extracellular matrix, making muscle fibers more susceptible to necrosis.
