Connective Tissue Growth Factor (CTGF) belongs to the CCN family of proteins, which consists of six members in humans: Cyr61, CTGF, Nov, WISP-1, WISP-2, and WISP-3. CCN proteins play a role in regulating cellular functions such as proliferation, differentiation, adhesion, and migration. They are expressed in various embryonic tissues and are involved in the development of organs like the kidney, nervous system, and bone. In adulthood, they are important for wound healing, bone repair, and can be implicated in diseases like cancer and fibrosis. Full-length CCN proteins have antiproliferative effects, while truncated isoforms may stimulate proliferation. The full-length CTGF protein consists of four modules with unique functions, including sequences that aid in cell adhesion and binding to glycoconjugates. Proteolysis of full-length CCN proteins can produce bioactive peptides, with some isoforms having oncogenic potential. Elevated CCN2 expression has been linked to various cancers, including breast and pancreatic tumors. Studies have also shown a correlation between CCN2 expression and tumor malignancy. In brain tumors, CCN2 has been associated with angiogenesis and cell proliferation. In leukemia, CCN2 expression was found in a higher proportion of mononuclear cells.
