Connective Tissue Growth Factor (CTGF) is a member of the CCN family of proteins, which includes six members in humans: Cyr61, CTGF, Nov, WISP-1, WISP-2, and WISP-3. These proteins are associated with the Extracellular Matrix (ECM) and cell membrane. CCN proteins play a role in regulating cellular functions such as proliferation, differentiation, survival, adhesion, and migration. They are expressed during embryonic development and play a role in various tissues including kidney, nervous system, muscle, bone marrow, cartilage, and bone.<br />Full-length CCN proteins exhibit antiproliferative activity, while truncated isoforms are likely to stimulate proliferation and act as oncogenes. The full-length CTGF protein consists of four modules, each with specific functions. Proteolysis of full-length CTGF proteins, such as CCN2 and CCN3, may result in the production of CCN-derived peptides with high affinity for ligands. Elevated expression of CCN2 has been detected in various pathological conditions, such as invasive mammary ductal carcinomas and pancreatic tumors. In brain tumors, CCN2 plays a role in angiogenesis and is associated with increased cell proliferation. Downregulation of CCN2 expression leads to growth arrest, while overexpression induces cell proliferation.<br />CCN2 expression has also been observed in mononuclear cells of patients with acute lymphoblastic leukemia.
