Type IV collagen, a crucial component of basement membranes, consists of three alpha subunits encoded by six different genes (alpha 1 through alpha 6). These subunits can combine to form a triple helix structure. In Goodpasture syndrome, autoantibodies target collagen molecules in alveoli and glomeruli basement membranes, specifically binding to the non-collagenous C-terminal domain of the protein. Unique alternative transcripts are found in the human COL4A3 gene, showing limited alternative splicing in the six exons that encode this C-terminal domain. COL4A3 is also associated with the autosomal recessive form of Alport syndrome, with mutations occurring in the exons that encode the C-terminal region. COL4A3 is organized in a head-to-head arrangement with another type IV collagen gene, both sharing a common promoter. Additionally, some exons of COL4A3 are interspersed with exons of an unidentified gene on the opposite strand.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
