Clusterin, also known as Apolipoprotein J (APO-J), is a 75-80 kD disulfide-linked heterodimeric protein enriched with sialic acid. Truncated forms that target the nucleus have been identified. The precursor polypeptide chain is cleaved proteolytically to remove the secretory signal peptide and generate two chains, a and b. These chains are assembled in an anti-parallel manner, forming a heterodimeric molecule. Clusterin shows a high degree of sequence homology across various species, ranging from 70% to 80%. It is expressed in most mammalian tissues and can be found in bodily fluids such as plasma, milk, urine, cerebrospinal fluid, and semen. Clusterin has the ability to bind and form complexes with numerous partners, including immunoglobulins, lipids, and bacteria. It has been associated with functions such as phagocyte recruitment, apoptosis inhibition, and lipid transport. The exact function of clusterin remains unclear, but one hypothesis suggests that it acts as an extracellular chaperone protecting cells from stress-induced insult caused by degraded proteins. Clusterin is known to be up- or down-regulated in various pathological conditions, including cancer, Alzheimer's disease, and autoimmunity.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
