CFI is an enzyme that plays a role in the inactivation of C3b and C4b. However, it needs a cofactor like factor H or C4b binding protein to become active. When CFI becomes active, it cleaves the alpha-peptide chain of C3b and C4b, resulting in the production of iC3b and iC4b. This cleavage process deactivates the functions of C3b and C4b in the complement system. Additionally, CFI can cleave the alpha chain of C3b two times, leading to the release of a small fragment called C3f. Furthermore, in the presence of CR1, CFI has the ability to cleave iC3b and separate C3c from C3dg. As for C4b, it is rapidly cleaved at two sites, which separates C4c from C4d.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
