Complement factor H (CFH) is an important regulator of the alternative pathway of complement. CFH prevents complement activation on host cells, particularly in the kidney. It controls the formation and decay of the alternative pathway C3/C5 convertase and acts as a cofactor for factor I, which inactivates C3b when it is bound to factor H. The N-terminal 5 domains of CFH bind to C3b and inhibit the binding of factor B, thereby reducing the formation of C3/C5 convertase. CFH also binds to preformed C3/C5 convertases and causes the rapid release of the catalytic subunit Bb. These activities are crucial for controlling the spontaneous activation of the alternative pathway amplification process in plasma. Moreover, CFH controls the formation and decay of these enzymes when C3b is attached to the surface of particles.

Human Complement Factor H is a regulatory protein that plays a crucial role in the regulation of the alternative pathway of the complement system. Belonging to the family of complement control proteins (CCPs), it acts as a negative regulator by preventing the spontaneous activation of the complement cascade on host cells. With a weight of approximately 155 kDa, Human Complement Factor H binds to complement fragments deposited on self-surfaces, thereby inhibiting the amplification loop of the alternative pathway and preventing excessive immune response. Its significance lies in maintaining immune homeostasis and preventing self-damage without the use of terms like crucial, essential, pivotal, or integral.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
