CD33 is a hypothetical adhesion molecule found on specific kinds of cells that arise from the myelomonocytic lineage. It plays a role in cell binding through a process that depends on the presence of sialic acid. Specifically, CD33 shows a preference for binding to sialic acid molecules that are connected by alpha-2,6-linkages. However, the area on CD33 that recognizes sialic acid is covered up by interactions with other sialic acid molecules on the same cell surface. In the immune response, CD33 acts as an inhibitory receptor. This occurs when ligands induce phosphorylation of tyrosine residues, which in turn leads to the recruitment of cytoplasmic phosphatases through their SH2 domains. These phosphatases obstruct signal transduction by dephosphorylating signaling molecules. Furthermore, CD33 promotes apoptosis in cases of acute myeloid leukemia.
