Calcium-activated nucleotidase 1 (CANT1) belongs to the apyrase family and acts as a calcium-dependent nucleotidase, displaying a preference for UDP. Its activity with different substrates follows this order: UDP > GDP > UTP > GTP. CANT1 has minimal activity towards ADP and ATP, and does not hydrolyze AMP and GMP. Although the specific function of CANT1 is not yet known, its substrates play a crucial role in various signaling functions, such as the release of calcium through the activation of pyrimidinergic signaling. Mutations in the CANT1 gene have been associated with Desbuquois dysplasia with hand anomalies.

This enzyme hydrolyzes nucleoside triphosphates and diphosphates, thereby modulating purinergic signaling pathways. Its precise functions and mechanisms are still being investigated, making hCAN1 an intriguing target for further research in various physiological and pathological contexts.
