Native human C8 is a glycosylated protein composed of three polypeptide chains: alpha, gamma, and beta. The alpha and gamma chains are connected by disulfide bonds, while the beta chain is non-covalently bound to the alpha-gamma complex. C8 plays a crucial role in the formation of the membrane attack complex (MAC) and is activated by binding to newly-formed C5b, C6, and C7 complexes on the cell membrane. During complement activation, proteolytic enzyme complexes are generated by each pathway, which then bind to the target surface. These enzymes cleave a peptide bond in the larger alpha chain of C5, resulting in the release of C5a and the activation of C5b. Although C5b is unstable, it remains bound to the activating complex for a few minutes, during which it can bind a single C6 molecule from the surrounding fluid or decay and lose its ability to form MAC. The C5b,6 complex can also remain connected to the C3/C5 convertase, and the binding of a single C7 molecule exposes a membrane-binding region. This allows C5b,6,7 to enter the bilipid layer of the target cell. Each C5b-7 complex has the ability to bind to one molecule of C8, which enhances its integration into the cell membrane.

Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
