C7 is essential for the formation of the membrane attack complex. It is activated when it binds to recently-formed C5b,C6 complexes at the cell membrane. Proteolytic enzyme complexes are produced by each pathway of complement activation, which bind to the target surface. These enzymes cleave a peptide bond in the larger alpha chain of C5, releasing C5a and activating C5b. Although C5b is unstable, it remains bound to the activating complex for a short period of time. During this time, it can bind to a single C6 from the surrounding fluid or decay and lose its ability to form the membrane attack complex (MAC). The C5b,6 complex can also remain connected to the C3/C5 convertase. When a single C7 binds to it, a membrane-binding region is exposed, allowing C5b,6,7 to enter the bilipid layer of the target cell.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
