C6 is an essential component in the formation of membrane attack complex (MAC). It becomes activated by attaching to newly-formed C5b on the cell membrane. Each pathway of complement activation creates enzyme complexes that are bound to the target surface. These enzymes cleave a peptide bond in the larger alpha chain of C5, resulting in the release of C5a and the activation of C5b. Although C5b is unstable, it remains attached to the activating complex for a short period of time. During this time, it can either bind a single C6 from the surrounding fluid or decay and lose its ability to form MAC. The C5b,6 complex can also remain connected to the C3/C5 convertase. When a single C7 binds to this complex, it exposes a region that can bind to the cell membrane, allowing C5b,6,7 to enter the target cell's bilipid layer.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
