C4BP

Alternative Names: C4bp, C4BP, C4b-binding protein alpha chain, Proline-rich protein, PRP, C4BPA

C4BP is a flower-like protein structure consisting of alpha chains radiating from a central core. The arms of C4BP are connected by disulfide bonds and can bind to C4b at their ends. This allows C4BP to act as a cofactor for factor I, facilitating the splitting and permanent inactivation of C4b. The main role of C4BP is to regulate complement activation in the classical and lectin pathways. By binding to C4b, C4BP accelerates the dissociation of C2a from the C3/C5 convertase C4b,C2a, ultimately deactivating the central enzyme of both pathways. Research has demonstrated that C4BP is capable of binding up to four C4b molecules simultaneously on surfaces densely covered with C4b.

Product Description

Human Complement Component 4 Binding Protein, also known as C4BP, is a glycoprotein that plays a significant role in the regulation of the complement system. Belonging to the family of complement control proteins, C4BP acts as a cofactor for the proteolytic inactivation of the complement protein C4b. With a molecular weight of approximately 570 kDa, this binding protein aids in preventing excessive activation of the complement cascade, thereby maintaining immune homeostasis. Through its interaction with C4b, Human Complement Component 4 Binding Protein modulates the immune response, serving as a regulatory component of the complement system.

Storage Instruction

Reconstituted store at -20°C or -80°C.We recommend that single use aliquots are prepared to avoid freeze-thaw cycles.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
