C3d is obtained from human C3 through a sequence of proteolytic cleavages. First, purified human C3 is converted to C3b using natural human C3 convertase. This C3b is then transformed into iC3b. Finally, trypsin is used to cleave iC3b and produce C3d. During complement activation on a surface, if the C3b precursor is attached, the iC3b, C3dg, and C3d fragments remain covalently bonded to that surface. Only during complement activation on a surface can C3d bind to a surface. The attachment of surface-bound C3d to the target is through an ester or an amide covalent bond. However, since ester bonds are unstable, C3d eventually detaches from the particle.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
