Native human C3a is produced through the cleavage of human C3 protein by a human C3 convertase. C3a belongs to the anaphylatoxin family comprising C3a, C4a, and C5a, which are formed upon complement activation. C3a is responsible for various inflammatory responses such as smooth muscle contraction, increased vascular permeability, vasodilation, and the release of histamine from mast cells and basophils. Unlike C5a, C3a does not have significant neutrophil stimulating effects and does not induce chemotaxis, granule release, or superoxide production. C3a exerts its effects by binding to the C3a Receptor, a G-protein coupled receptor found primarily in peripheral tissues, lymphoid cells, and in the central nervous system. The activities of C3a can be deactivated by the removal of the C-terminal arginine residue, which occurs rapidly in plasma through the action of carboxypeptidase N.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
