Native human C3 is a glycosylated polypeptide with two disulfide-linked chains. C3 plays a central role in activating all three pathways of complement activation. Each pathway initiates the formation of proteolytic enzyme complexes that bind to the target surface. These enzymes cleave a peptide bond in C3, resulting in the release of C3a (an anaphylatoxin) and activation of C3b. However, most activated C3 does not bind to the surface. Instead, it reacts with water to form fluid phase C3b, which is rapidly inactivated by factors H and I to form iC3b. Surface-bound C3b is essential in all three pathways for efficient activation of C5 and the formation of C5b-9 complexes, which ultimately lead to the lysis of the target cell membrane.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
