C1r is the active form of the C1r proenzyme in the C1 complex of the complement system. C1 is a calcium-dependent complex comprised of 1 C1q, 2 C1r, and 2 C1s molecules. C1q binds to the Fc domains of IgG or IgM using multiple arms, activating C1r to cleave and activate the C1s protease zymogens. Cleavage of C1r results in two fragments of 57,000 and 35,000 Dalton.

Human Complement C1r is a serine protease that belongs to the family of complement system proteins. It plays a key role in the activation of the classical complement pathway by cleaving C1s, leading to the formation of the C1 complex. Complement C1r has a molecular weight of approximately 80 kDa and is involved in the immune response against pathogens and foreign substances. Its enzymatic activity is tightly regulated to prevent excessive activation of the complement system.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
