C1q is a component of the classical pathway of complement activation. It forms the C1 complex with the enzymatically active components C1r and C1s. When C1 binds to immunoglobulins in the form of immune complexes, it activates C1r and C1s proteases and further activates the classical pathway of complement. C1q is a glycoprotein that belongs to the collectin family. It has a molecular weight of about 410-462 kDa and is a hexamer composed of globular heads attached to collagen-like triple-helix tails. The globular heads of C1q bind exclusively to the CH2 domain of IgG or the CH3 domain of IgM. Each heavy chain of the immunoglobulin molecule has a single binding site for C1q. For C1q to activate and change conformation, it must bind to at least two heavy chains in the form of immune complexes attached to multivalent antigens. The main role of C1q is to clear immune complexes and apoptotic bodies from the body. Disruption of this process can lead to the development of autoimmunity. Genetic deficiencies of C1q or other components of the classical pathway increase the risk of developing systemic lupus erythematosus (SLE). C1q specifically binds to apoptotic bodies of human keratinocytes, vascular endothelial cells, and lymphocytes and plays a role in their clearance. This helps prevent the stimulation of the immune

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
