P85a acts as the regulator for class IA PI3-kinase isoforms a, b, and d. It has two SH2 domains that bind to tyrosine-phosphorylated growth factor receptors or substrate adaptor proteins. P85a also contains a BH domain with GAP activity towards small GTPases Rab4, Rab5, Cdc42, and Rac1. Research has shown that phosphorylation of the p85a adapter by the PI3Ka catalytic subunit reduces the lipid kinase activity of the heterodimer, suggesting PI3K-dependent signaling events may not require production of 3’-phosphorylated phosphoinositides. The protein kinase activity of PI3Ka is involved in IRS-1 serine phosphorylation in insulin-treated adipocytes and in STAT3 and IRS-1 phosphorylation upon activation of the type 1 IFN receptor by IFN-a.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
