BAG3 is a protein that inhibits the chaperone activity of HSP70/HSC70 by promoting the release of substrates. It has anti-apoptotic properties. BAG proteins, including BAG3, interact with Hip to bind to the ATPase domain of Hsc70/Hsp70 and facilitate substrate release. BAG proteins have a BAG domain close to the C terminus, but they differ in their N-terminal regions. In the case of BAG3, it has a WW domain in the N-terminal region and a BAG domain in the C-terminal region. In vitro and in mammalian cells, the BAG domains of BAG1, BAG2, and BAG3 specifically bind with high affinity to the Hsc70 ATPase domain and inhibit its chaperone activity. BAG3 acts as a protein-refolding cochaperone of the bcl2 binding protein BAG family and is upregulated in response to persistent cellular calcium imbalance. It has been shown to reduce stress-induced apoptosis.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
