Zinc-alpha-2-glycoprotein (ZAG) is present in body fluids like serum and sweat, as well as in seminal and breast cyst fluids. It shares the same amino acid sequence as lipid mobilizing factor (LMF), which is associated with the significant loss of adipose body stores in cancer cachexia. ZAG has been shown to stimulate the breakdown of fat cells both in living organisms and in laboratory settings. It has been suggested that ZAG may play a role in regulating body weight, age-related changes in obesity, and melanin production by melanocytes. ZAG is classified as a novel adipokine because it is produced by white and brown fat cells and may act on fat reduction in cachexia. Understanding ZAG's function in normal body fat regulation is important. ZAG has a similar structure to class I major histocompatibility complex (MHC) proteins but is not attached to cell membranes and does not associate with alpha-2-microglobulin in humans. Similar to MHC proteins, ZAG has a groove at the top, and x-ray crystallography has revealed a mysterious element in the same position as antigenic peptides in MHC proteins and glycolipids in CD1 isoforms. The unidentified substance is likely not a peptide and cannot be a glycolipid due to the groove's size.
