ATPase inhibitory factor 1 (ATPIF1) regulates the activity of F1-ATPase, which is responsible for energy production in cells. ATPIF1 attaches to a specific region of the F1-ATPase and interferes with the interaction between its beta and gamma subunits. This control mechanism is particularly relevant at low pH levels. In addition to this role, ATPIF1 is involved in the development of cancer. Overexpression of ATPIF1 in human carcinomas contributes to oncogenesis and provides insight into the altered energy metabolism observed in cancer cells. ATPIF1 also acts as an endogenous inhibitor of F1F(o)-ATPase, preventing excessive depletion of ATP when the mitochondrial membrane potential decreases below a certain threshold. This happens when the F1F(o)-ATP synthase starts hydrolyzing ATP to pump protons out of the mitochondrial matrix.

Sample Prep Instruction

1. Thaw the vial on ice, spin briefly
2. Prepare single-use or stock aliquots. This stage will depend on your final application so please adapt as appropriate. All our proteins are supplied carrier protein-free. If compatible with your work and you are storing at lower concentrations (<50 ug/ml) adding carrier protein is highly advised (usually 1% w/v high purity BSA or equivalent – ensure all buffers are sterile-filtered).
3. Prepare single-use aliquots whenever possible to avoid freeze-thaw cycles which can damage the proteins and reduce bioactivity. Store aliquots at -70 °C (or -20 °C)
4. We recommend sterile filtering after dilution in media or the final working solution
